#N rat, pituitary, LH, immunocytochemistry, ontogenesis, proliferation
#T Materials & Methods: Rat fetuses (17.5-21.5 days of gestation) and
 neonates (3, 7, 14 and 28 days) were used. Bromodeoxyuridine (BrdU,
100mg/KgBW) was i.p. injected to the mother rat or neonates 2 hours
prior
to sacrifice and the pituitaries were fixed with Bouin's fluid.
Paraffin sections were consecutively immuno-stained with anti-BrdU and
anti-ovine LHb and
the number of positive cells were counted.
Results: The cells labeled with BrdU were around 600-800/mm2
through the fetal period and 3 days after birth and then gradually
decreased to 146/mm2 at 28 days. The cells labeled with anti-LHb were
first
detected at 18.5 days and remained below 100/mm2 throughout the fetal
period but they rapidly increased after birth to reach 800/mm2 at 28
days.
The cells double-labeled with both anti-sera were detected at 3-14 days
after birth. But they comprised only 3% or less of total gonadotrophs
so it
seemed that the majority of go-nadotrophs were derived from
differentiation of some kind of stem cells with little contribution by
mitoses of existing
gonadotrophs.
