#N Golgi apparatus, beta-COP, brefeldin A, okadaic acid, parotid
gland, rat
#T Double-label immunocytochemistry of beta-COP and mannosidase II
(man II) was applied in the parotid acinar cells after the treatment with
Golgi-disrupting agents. In untreated acinar cell, immuno-reactivity of
beta-COP and man II mainly associated with the Golgi apparatus. Brefeldin
A (BFA) caused rapid redistribution of man II into the endoplasmic
reticulum (ER) accompanied with vesicula-tion of the Golgi stack. beta-COP
lost its association with the Golgi region to disperse throughout the
cytoplasm. Okadaic acid (OA) also induced rapid vesiculation of stacked
structure of the Golgi apparatus, but the redistribution of man II into the
ER could not be observed. Of interest is that beta-COP markedly
accumulated to the region of the disrupting Golgi apparatus on which man II
immuno-reactivity has been preserved. These results suggested that BFA has
an inhibitory effect on the attachment of beta-COP to membranes, resulting
in the prevention of transport vesicle formation. In con-trast, OA seemed
not to prevent the formation of vesicles but inhibit the fusion of them to
the target membranes. As a result, beta-COP-coated membranes failed to be
integrated into Golgi apparatus, and were accumulated in the Golgi region.
